Abstract
We have been developing a technology called Upcyting. This makes it possible to propagate human primary cells directly and efficiently in cell culture without using stem cells. A second approach is to “upgrade” liver cancer cell lines such as HepG2 with relevant enzymes involved in biotransformation. Our aim is to generate and use physiologically relevant human cells such as hepatocytes for basic cell biology research and drug testing, including cytotoxicity studies. An important research focus of our lab is the investigation of the activity of phase I and phase II enzymes in hepatocytes with particular regard to the formation of drug metabolites. We are also investigating potentially toxic effects of chemicals on human cell culture systems using in vitro toxicology readouts such as metabolic activity, apoptosis, FACS and double strand break formation (in case of drugs with genotoxicity).
