蛋白质聚集及其抑制

Protein Aggregation and its Inhibition

 Misfolding and fibrillar aggregation of proteins result in a lot of serious disorders, such as Alzheimer’s disease, the main hallmark of which is the fibrillogenesis of amyloid β-protein (Aβ) into plaques in the brain. Hence, inhibition of protein aggregation is one of the major concerns in life and medical sciences. We have made efforts on the discovery and design of potent inhibitors against the amyloid as well as metal-mediated amyloid toxicity, including discovery of natural compounds, design of peptide-based inhibitors, and development of nano-inhibitors and protein-based inhibitors. This talk starts with an introduction of our finding of a new inhibition phenomenon that is explained by Hydrophobic Binding-Electrostatic Repulsion effect (HyBER effect), and the design of inhibitors based on the HyBER theory. This is followed by our efforts to design peptide-based inhibitors and their conjugations to enhance their performance and/or to introduce new functionalities. Finally, our recent studies of carbon dots as a potent scavenger and detector targeting Aβ plaques as well as the enhancing effect of near-infrared light-driven nanomotor on the inhibitory efficiency of a peptide inhibitor are discussed.

1983年天津大学化学工程系本科毕业,1990年获得东京大学工学博士学位。 199311月任天津大学化学工程系教授。1996年获得国务院政府特殊津贴1998年入选国家百千万人才工程第一、二层次人选,2000年获得国家杰出青年科学基金。国务院学位委员会第五、六届化工学科评议组成员Journal of Chromatography A主编,Biochemical Engineering JournalChinese Journal of Chemical Engineering副主编。

主要从事生物化工和生物医药工程研究,研究方向包括生物分离工程、生物催化、蛋白质折叠复性和蛋白质聚集及其抑制等。发表SCI收录论文330余篇,导毕业博士60余名,硕士100余名。指导博士生获得全国百篇优秀博士论文奖”1名、提名奖1名。获省部级自然科学一等奖1项,二等奖2项,亚洲生物技术联合会亚洲青年生化工程师学会奖1项。
